花刺参粘多糖对大鼠血管平滑肌细胞粘附分子表达的影响

【摘要】 目的:观察花刺参粘多糖对肿瘤坏死因子α诱导的大鼠血管平滑肌细胞血管细胞粘附分子1、细胞间粘附分子1表达的影响,以探讨花刺参粘多糖抗动脉粥样硬化的可能机制。方法: 组织贴块法体外原代培养大鼠血管平滑肌细胞,采用流式细胞术、逆转录-聚合酶链反应和细胞粘附实验方法,观察花刺参粘多糖对肿瘤坏死因子α诱导的平滑肌细胞血管细胞粘附分子1、细胞间粘附分子1蛋白及mRNA水平表达及单核细胞与平滑肌细胞粘附的影响。结果:肿瘤坏死因子α (10μg/L)刺激后可使血管细胞粘附分子1、细胞间粘附分子1表达显著增强,而花刺参粘多糖 (1.5g/L-6.0g/L)可从蛋白和mRNA水平浓度依赖性地抑制其表达,减少单核细胞与平滑肌细胞粘附(P<0.05)。结论:花刺参粘多糖可抑制肿瘤坏死因子α诱导的血管细胞粘附分子1、细胞间粘附分子1的表达,从而减少单核细胞与血管平滑肌细胞粘附和聚集,这可能对减轻血管壁的炎症反应、延缓动脉粥样硬化发生发展起到一定作用。

【Abstract】 Objective: To investigate the effects of Stichopus Variegates on tumor necrosis factor-alpha (TNF-α)Stichopus Variegates induced expression of vascular cell adhesion molecule-1(VCAM-1) and intercellular adhesion molecule-1(ICAM-1) in rat smooth muscle cells(VSMC) and to explore the mechamism of anti-atherosclerosis by Stichopus Variegates. Methods: SMC were isolated from the media of normal rat thoracic aorta, the VCAM-1 and ICAM-1 mRNA content and surface expression was detected by flow cytometric technique and reverse transcription-polymerase chain reaction. Adhesion of monocytes to SMC was measured by rose Bengal staining assay. Results: Under basal conditions of cultivation, both mRNA and membrane expression of VCAM-1、ICAM-1 were low .Treatment of SMCs with TNF-α(10μg/L) led to an increase in both VCAM-1、ICAM-1 mRNA and cell surface expression. Stichopus Variegates(1.5g/L-6.0g/L) significantly inhibited TNF-alpha-induced expression of each adhesion molecule, and effectively reduced monocytes adhesion to SMC (P<0.01) ; These inhibitory effects were dose-dependent. Conclusion: Stichopus Variegates inhibited TNF-α-induced expression of VCAM-1、ICAM-1 and monotyces-SMC adhesion, this medhanism may contribute to the anti-atherosclerotic role of Stichopus Variegates.
