LC-MS法测定大鼠血浆中槲皮素代谢物

【摘要】 目的采用液相色谱-质谱联用法鉴定大鼠口服槲皮素单体后血浆中的代谢产物。方法血浆样品经过2M盐酸(甲醇-水溶液)水解后,进行色谱分离,采用液相色谱-质谱联用仪检测,大气压电喷雾离子源(EPI)负离子条件下进行扫描,鉴定大鼠给药槲皮素(100mg·kg-1)单体后血浆中代谢产物的结构。结果通过与标准物紫外谱图比较,结合质谱仪扫描结果分析,在大鼠给药槲皮素(100mg·kg-1)单体后血浆中鉴定出槲皮素(m/z→301.0)和异鼠李素(m/s→314.9)两种代谢产物结构。结论采用盐酸酸水解的方法可快速、有效地鉴定出大鼠血浆中槲皮素代谢产物,为进行含槲皮素中药及其复方的药物代谢动力学研究奠定科学基础。

【Abstract】 Aim To determinate the metabolites of quercetin in rat plasma after an oral administration by Liquid Chromatography-Mass Spectrometry(LC-MS).Methods The rats were administrated quercetin monomer solution (100mg·kg-1)by oral administration and the blood sample were prepared.Quercetin and isorhamnetin were extracted from rat plasma after acid hydrolysis using 2M HCl in aqueous methonl.After acid hydrolysis,the analytes of interest were separated on a SUPELCOSILTMC18 column.The mobile phase was Methonl- Acetonitrile-1% Acetic acid(20:30:50)with a flow rate of 0.8mL·min-1.A Braker Mass Spectrometry equipped with an Atmospheric Pressure Ionization-Electrospray(EPI) source was used as detector and was operated in negative ion mode.Results Quercetin and isorhamnetin were identified through comparing with the standard spectrum and the results of mass spectrometry,i.e.m/z→301.0 was quercetin and m/z→314.9 was isorhamnetin.Conclusion Acetic acid hydrolysis was a fast feasible method and could be used to identify the quercetin plasma metabolites in rat.And this method could be acted as referring during pharcokinetics studies upon Chinese material medica.
