用抑制性消减杂交法分离兴春椪柑干旱胁迫诱导表达的cDNA

【摘要】 对兴春椪柑进行干旱胁迫,以其干旱胁迫叶 cDNA 为试验方(tester),正常生长的兴春椪柑叶 cDNA 为驱动方(driver),利用抑制性消减杂交技术(SSH)构建了干旱胁迫下兴春椪柑的消减文库。重组克隆经 PCR 检测,插入片段大部分集中在250~700 bp 之间.通过对文库阳性克隆随机挑取85个测序,成功获得79条 EST,获得非重复 EST 58条.
【Abstract】 A suppression subtractive hybridization library of Ponkan leaves(C.reticulata)was constructed by using the drought treated leaves as tester and leaves grown under normal conditions as driver.The PCR results of recombinant clones showed that the inserts were mainly 250~700 bp.A total of 85 positive clones were randomly picked out from the subtracted library and sequenced,a total of 79 positive clones were obtained,and 58 unique ESTs were gained bassed on the sequening data.
