5-氮杂胞苷对乳腺癌细胞(Bcap-37)中RASSF1A基因甲基化及其表达的影响

【摘要】 目的:探讨5-氮杂胞苷对人乳腺癌细胞Bcap-37细胞增殖的抑制作用及作用机制。方法:不同浓度的5-氮杂胞苷与Bcap-37细胞共培养后,应用细胞计数法检测Bcap-37细胞生长的情况;应用甲基化特异性PCR(MSP)检测5-氮杂胞苷前后RASSF1A基因甲基化、非甲基化状态的改变;采用逆转录PCR检测RASSF1A基因的mRNA表达。结果:当5-氮杂胞苷>5.00μmol/L剂量时对细胞生长呈浓度依赖抑制作用(P<0。01);5-氮杂胞苷作用4d后,对RASSF1A基因有去甲基化作用,细胞Bcap-37的RASSF1A基因mRNA表达明显增强。结论:特异性甲基转移酶抑制剂5-氮杂胞苷能较好地逆转Bcap-37细胞RASSF1A基因异常甲基化,诱导RASSF1A基因的表达,从而抑制肿瘤细胞生长
【Abstract】 Purpose:Inhibitory effect of 5-azacytidine on the proliferation of human breast cancer cell line Bcap-37 and its mechanism was investigated.Method: After treatment with 5-azacytidine for Bcap-37 cells,the cells proliferation was analyzed by cell counting.The methylation and unmethylation status of RASSF1A(RAS association domain familylA)gene were detected by methylation special PCR(MSP)and RASSF1A mRNA expression was detected by RT-PCR..Result:After treatment with 5-azacytidine(>5μmol/L),the Bcap-37 cells growth rate was decreased in a dose dependent manner;the demethylation of RASSF1A gene occurred and RASSF1A mRNA expression was incressed.Conclusion:5-Azacytidine may effectively enhance RASSF1A gene demethylation and enhance the expression of RASSF1A mRNA and inhibite the cell growth.
