慢性氟中毒砷中毒及联合中毒对小鼠心脏的氧化应激作用

【摘要】 目的探讨慢性氟中毒、砷中毒及氟砷联合中毒对小鼠心脏氧化应激作用和对G6P含量及SDH活性的影响。方法昆明种小白鼠120只,随机分成对照、氟中毒、砷中毒和氟砷联合中毒四组,分别自由进食普通、高氟、高砷和高氟高砷饲料,饲养12个月。分别检测心脏GSH含量、SOD和GSH-PX活性、脂质过氧化产物含量。结果发现三种中毒均引起心脏LPO含量显著升高P<0.01,心脏GSH含量及SOD和GSH-PX活性显著降低P<0.01,且以砷组和氟砷组为最低P<0.05;三实验组均显示SDHase和G6Pase活性下降(P<0.01),而砷组和氟砷组下降最为显著(P<0.05)。结论慢性氟中毒、砷中毒及氟砷联合中毒均引起小鼠心脏中LPO含量升高、抗氧化能力降低;慢性砷中毒和氟砷联合中毒引起细胞代谢异常;并且氟、砷两毒物对这些作用存在一定的相互影响。
【Abstract】 Objective To compare the effects of oxidation stree in mice heart caused by chronically exposed to fluoride,arsenic and fluoride-arsenic respectively.Methods 60 mices were randomly divided into four groups which were fed with four different diets supplemented with fluoride,arsenic and fluoride-arsenic respectively.After twelve months,the animals were sacrificed and LPO and GSH contents,the activities of SOD,GSH-PX,SDHase and G6Pase in the heart of intoxicated animals were determined.Results In whole three poisoned groups,LPO contents were significally increased,GSH contents,activities of SOD and GSH-PX were markedly decreased in defferent degree,and among which,two indece,SOD and GSH-PX of fluoride group showed higher than that of other two poisoned groups,.Furthermore,the activities of SDHase and G6Pase in three poisoned groups were different from that of control group with statistically significance. And the two indices of fluoride group displayed much higher than that of the other two poisoned groups. Conclusion In mice,the increase of LPO content and the decrease of antioxidation levels can be found in mice chronically fed with fluoride,arsenic or combining with fluoride and arsenic respectively.The chronical fluorosis,arsenism and fluoride-arsenic coalescent poisoning induced the abnormalities of cell metablism.To some extent,fuoride and arsenic can influence each other the these effects.
