基因枪介导的花生条纹病毒壳蛋白基因转化花生研究

【摘要】 用中花3号花生胚性愈伤为受体,通过基因枪介导将花生条纹病毒壳蛋白基因(PStV CP)和潮霉素基因(Hyg~r)同时导入花生体胚。轰击胚经潮霉素筛选2~3月得到转化胚,其基因枪转化率为0.4%~2.9%。转化胚再生成花生植株。PCR检测PStV CP和Hyg~r均导入花生基因组中,两基因共转化率为50%。转基因花生农艺性状无明显改变,对PStV抗性表现不显著。这是国内通过基因枪介导获得转基因花生的首次报道。

【Abstract】 Coat protein (CP) gene of peanut stripe virus (PStV) and Hyg~r gene were cotransferred into embryogenic callus produced from mature seed of peanut variety Zhonghua No. 3 by particle bombardment. The embryos produced from died embryogenic callus after 2~3 months selection of bombarded callus on MS medium containing 20mg/L hygromycin B were regenerated into peanut plants. Frequency of transformation was 0.4%~2.9%. PCR test of To generation plants indicated the PStV CP gene and Hyg~r have been inserted into peanut genome. Transgenic peanut plants were normal in morphology and fertile. The resistance level of transgenic plants to PStV has no significant increment compared with non-transgenic plants by virus inoculation.
