基于80C196KC20的变温气流式PCR仪

【摘要】 PCR是基因扩增所需经历的一种生物化学反应,而此反应成功与否的关键因素之一就在于必须满足其反应实体所需的温度循环条件即高温变性,低温退火,中温引物延伸,每一阶段都有其具体的时间要求。选用变温气流方案,采用80C196KC20单片机系统通过控制PWM输出的占空比,对固态继电器的通断进行控制,进而控制电热丝加热功率从而达到对气流介质控温的目的。此方法变温迅速,易于设定不同的变温程序,可以满足不同的试验需求。
【Abstract】 The realization of gene amplification is based on a biochemical reaction named PCR. One of the key factors which affect the result of the reaction is to meet its condition of temperature circle. A temperature circle ends after metatropy, annealing and primer extension. The three stages take place in turn under high, low, medium temperature and each stage must be finished in concrete time. A poikilothermal airstream scheme is adopted and a 80C196KC20 microcomputer system is used to control the output of the pulse duration ration of PWM wave to control airstream’s temperature through solid state relay. This method enables us to get rapid alternating temperature and to develop different programs easily so that it can meet different experimental demands of gene amplification.
