深圳哈萨克族移民DNA氧化损伤的分子流行病学研究

【摘要】 目的比较研究来深圳不同年份的新疆哈萨克族居民的 DNA 氧化损伤作用,从而探讨移民对 DNA 氧化损伤的影响。方法选取60个深圳的新疆哈萨克族居民,按来深圳不同年份分为3组(每组20人),调查其移民后主要生活方式的变化情况;采静脉血做彗星实验,收集24h 尿测定8-OH-dG。结果彗星实验显示,来深圳3~年组 DNA 损伤程度比~1年组均显著降低 (R<0.05);8-OH-dG 测定结果显示,移民1~3年组的8-OH-dG 水平低于~1年组(P<0.05),移民3~年组的8-OH-dG 水平明显低于~1年组(P<0.01)。结论移民的相关生活方式变化可能具有降低哈萨克族居民的 DNA 氧化损伤的作用。
【Abstract】 Objective To explore the relationship of migration and oxidative DNA damage by comparative study of oxidative DNA damage effects of different migration years of Xinjiang Kazak ethnic migrants in Shenzhen.Methods Sixty Kazak ethnic people in Shenzhen were selected,and were divided into three groups (n=20) according to their migration years,the major changes of their life style were investigated.8-hydroxy-2’-deoxyguanosine(8-OH-dG) levels in urine were analyzed,and comet assay of peripheral blood lymphocytes was conducted.Results Compared with migration~1 year group,a significant decrease of oliveive tail moment and tail/head length in comet assay in migration 3~years group was observed (P<0.05).The 8-OH-dG level decreased significantly in migration 1~3 years group (P<0.05) and migration 3~years group (P <0.01).Conclusion The results suggested that life style changes related to migration wight reduce DNA damage in Hasake nationalities.
