荧光差异显示技术在Bt上的初步应用

【摘要】 初步将荧光差异显示技术(Differential fluorescence induction)应用于苏云金芽胞杆菌(Bacillus thuringiensis,简称Bt)研究;提取了库斯塔克亚种菌株8010(Bt subsp.kurstaki 8010)高质量的总DNA,并利用Sau3AⅠ将其部分酶切,获得集中于0.5-3kb左右目的片段条带,构建pHT315gfp启动子荧光捕获载体;获得合适的基因组部分酶切条件,并构建了一个启动子荧光捕获载体;初步将荧光差异显示技术应用于Bt功能基因组学研究。
【Abstract】 Precliminary application of differential fluorescence induction technique on Bacillus thuringiensis(Bt).Total DNA of Bt subsp.kurstaki 8010 with high concentration and good quality was obtained,and the DNA was then partially digested by Sau3A I to recover 0.5-3 kb fragments.A promoter fluorescent trapping vector pHT315gfp was also constructed.The partially digestion condition was estabished and a promoter fluorescent trapping vector was obtained.These works may lay an important foundation on the application of differential fluorescence induction on B.thuringiensis functional genomics.
