红菜薹游离小孢子培养和植株再生的关键因素研究
Study on the key-factors to isolated Microspore Culture in purple flowering stalk (Brassica campestris L.ssp.Chinesis var.purpurea Tsen et Lee)

【摘要】 <正>对影响红菜薹游离小孢子培养和植株再生的关键因素进行了研究。结果表明:在小孢子胚诱导上,22个基因型之间的小孢子胚诱导率差异显著:对供体材料低温处理,在0~5天范围内小孢子胚诱导率差异不大,超过5天则明显降低;高温诱导在12~60h范围内差异不大,超过此范围则小孢子胚诱导率明显降低:NLN培养基中活性炭的有无和浓度大小对小孢子胚诱导率影响极大;在植株再生方面,对胚进行低温处理和提高琼脂浓度能提高胚植株的再生率。
【Abstract】 Several factors affecting isolated microspore culture in purple flowering stalk (Brassica campestris L.ssp.Chinesis var.purpurea Tsen et Lee) were studied.The results showed that there were significant differences in the frequency of microspore-induced embryos among different genotypes.For the low temperature treatment,no significant differences had been found during 0-5 days,while the frequencies were obviously lower when the period was longer than five days;there were no significant differences in high temperature when the incubation time was within 12-60h,but the frequencies of microspore-induced embryos were much lower over this duration;the presence and concentration of activated charcoal in NLN medium had great influence on the frequency of microspore-induced embryos. To the embryos produced from microspore culture,the regeneration frequency was heightened while treated with low temperature or solidified with high concentration of agar in the medium.
