与黄瓜矮生基因连锁的ISSR标记
An ISSR Marker Linked to the Dwarf Gene of Cucumber

【摘要】 以黄瓜品种D8(矮生)×JIN5(蔓生)的124株F2代为试材,采用ISSR的分子标记技术和BSA(混合群体分组分析法)法寻找与黄瓜矮生基因紧密连锁的分子标记。在80条ISSR引物的筛选中,引物UBC818扩增得到一条黄瓜矮生性状的多态性片段,经过F2代单株验证,此片段能在试验所用材料的蔓生植株中稳定出现,而在矮生植株中不出现。
【Abstract】 124 plants of F2 population from the cross combination of the dwarf line D8 and the long line JIN5 were used in this study to screen the molecular marker related to the dwarf gene of cucumber with ISSR marker and BSA(bulked segregation analysis) method.UBC818,one out of eighty ISSR primers,was screened to present polymophism specified in the dwafr line.The marker from the result of PCR with UBC818 was testified with individual DNA of the F2 population and the band could only be amplificd in the long vine plants,but not in the dwarf plant.
