农杆菌介导花生白藜芦醇合酶基因转化马铃薯的研究

【摘要】 利用pCAMBIA1301质粒构建的花生白藜芦醇合酶基因(Genbank AY170347) 植物表达载体pB3RS,导入农杆菌,用利福平和卡那霉素筛选并经PCR检测得到含有重组质粒的农杆菌菌株,以马铃薯“郑薯6号”试管薯切片为受体材料,转化马铃薯,从中筛选出潮霉素抗性植株,部分抗性植株经PCR检测检出目的条带。
【Abstract】 pCAMBIA1301 was utilized to construct the plant expression vector pB3RS carrying peanut resveratrol synthase gene and transformed to Agrobacterium Tumefaciens LBA4404. Agrobacterium Tumefaciens with the recombinant expression plamid was screened by kanamycin and rifampincin and confirmed by PCR. Potato "ZhengShu No. 6" was used as the transformation receptor and hygromycin - resistant transformants were obtained. PCR showed that the foreign gene existed in some hygromycin - resistant transformants.
