小果型西瓜同源四倍体的诱导及其特性研究

【摘要】 采用秋水仙素滴苗处理和未成熟子叶离体培养等方法,对6个小果型西瓜品系和1个大果型品系进行了同源四倍体的诱导及其特性研究。结果表明,在两片子叶夹角约160°时,去掉露头的小真叶,用0.2%的秋水仙素滴生长点4 d,诱变效果最好,7个品系的平均诱导成功率为13.57%,最高的达到32.5%,分别是常规滴苗处理的4.78倍和4.85倍。未成熟子叶离体培养诱导四倍体的适宜秋水仙素浓度和处理时间是0.05%和5 d,诱变成功率为23.3%。四倍体的小果型西瓜生长势强,单瓜重与二倍体的差异不大,固形物含量略有提高,结籽率显著降低。

【Abstract】 The induction of tetraploid watermelon for six diploid watermelon varieties with small fruits and one with big fruits was conducted by different methods . The results showed as follows ? when the small leaves of seedlings were peeled off and the bare growth tip was treated with 0.2% colchine for 4 days, the mean frequency of tetraploid plants obtained from seven varieties was about 13. 57%,and the highest frequency was 32. 5%,which were 4. 78 times and 4. 85 times that of CK(without peeling off the small leaves) respectively. The suitable colchicine concentration and treatment time were 0. 05% and 5 days respectively for inducing tetraploid of immature embryo in vitro, and the frequency of tetraploid plants was 23. 3%. The more strong growth potential displayed in the tetraploid watermelon plants than that of the diploid. The size of fruits without obvious difference between diploid and tetraploid, and the solid substance of fruit flesh increased slightly in the tetraploid, but the seed setting reduced obviously in the tetraploid.
