花椰菜花药培养研究
Study on Anther Culture of Cauliflower

【摘要】 以5个花椰菜品种为材料,进行花药离体培养。结果表明愈伤组织的诱导、芽的分化频率与供试材料基因型、低温预处理、培养基中蔗糖浓度和激素配比有关。对大多数基因型来说,适合诱导花药愈伤组织的培养基是MS+3%蔗糖+2,4-D 0.5 mg/L+BA 1.0 mg/L,愈伤组织分化率都达到40.6%以上。诱导花椰菜花药愈伤组织芽分化较好的培养基为MS+3%蔗糖+NAA 0.2 mg/L+BA 2.0 mg/L。再生植株炼苗后成功地移栽到了田间,并获得了自交种子。
【Abstract】 Five cauliflower (Brassica oleracea var. botrytis) cultivars’ anther were in vitro cultured in different media. The result showed that the induction of callus , differentiation of bud were related to genotype, low temperature-pretreatment, sucrose strength, and combination of growth regulators. In generally, it is better for callus induction that the anther were cultured in MS+3%sucrose+0. 5 mg/L 2,4-D+1. 0 mg/L BA, the rate of callus induction was more than 40. 6%, the medium, MS+3%sucrose+0. 2 mg/L NAA+2. 0 mg/L BA, was better for the bud differentiation. The regenerated plantlets were transplanted to field and the inbred seeds were obtained.
