白菜细胞核雄性不育相关基因CYP86MF的克隆和序列分析

【摘要】 用mRNA差别显示技术对白菜核雄性不育两用系中不育株群与可育株群花营总RNA进行了比较分析。在6种反转录模板和20种差异随机引物共120种组合的选择扩增中共回收了18个cDNA差异片段,经Northern验证获得了在可育株群中特有的并在花蕾中特异表达的一阳性差异片段,简称为B0302-4。B0302-4片段经3’RACE和5’端序列PCR扩增,获得了完整的3’PolyA尾和从起始密码子开始的5’序列,全长1667 bp,与拟南芥第—条染色体上编码细胞色素P450基因CYP86C4核苷酸序列有86%同源性,氨基酸同源率为83%。
【Abstract】 The difference was compared by mRNA differential display technology in Chinese cabbage pak-choi (Brassica campestris L. ssp. chinensis Makino var. cammunis Tsen et Lee) flower bud among genie male-sterile (GMS) line (A line) and maintainer line (B line). 18 differential fragments have been obtained in total of 120 selective amplifications. A differential fragment named as B0302-4 was confirmed by Northern hybridization. B0302-4 was shared by a maintainer line, and B0302-4 expressed specially in flower bud. The gene sequences 3’ side and 5’ side were identified by 3’ RACE and 5’ side PCR amplification. The gene is composed of 1667 base pair, and has 86% nucleotide similarity to a gene CYP86C4 encoding cytochrome P450 of Arabidopsis, and has 83% amino acid similarity to CYP86C4.
