黄皮花粉生活力测定方法的对比研究

【摘要】 采用花粉离体萌芽法和4种不同的染色法对普通黄皮、鸡心黄皮、无核黄皮、白糖黄皮的花粉生活力进行了测定,并对影响黄皮花粉生活力的培养条件及测定黄皮花粉生活力的4种染色法进行了分析比较。研究结果表明,普通黄皮和鸡心黄皮的花粉发芽率在培养基蔗糖浓度分别为15.0%和10.0%、无核黄皮和白糖黄皮在5.0%时培养效果最高。不同品种、培养基蔗糖浓度对花粉管平均长度的影响差异极显著,无核黄皮的花粉管平均长度为其他3个供试品种的1/2,不同染色方法在不同黄皮品种花粉生活力的测定结果上也存在极显著差异。
【Abstract】 The pollen activity of Common Chinese wampee, Chicken heart Chinese wampee,Seedless Chinese wampee and White sugar Chinese wampee were determined by detached pollen germination method and four kinds dyeing method. The culture condition which influenced the pollen activity of Chinese wampee and the four kinds of different dyeing methods which used to determine the pollen activity of Chinese wampee were analyzed. The result shows that the culture medium which contained 15. 0% and 10. 0% sucrose is the best culture medium to the pollen germinate rate of Common Chinese wampee and Chicken heart Chinese wampee. The culture medium which with the highest pollen germination rate of Seedless Chinese wampee and White sugar Chinese wampee is culture medium which contained 5. 0% sucrose. The difference on the pollen activity of Chinese wampee which caused by different species and different culture medium was at the level of extreme significance. Common Chinese wampee,Chicken heart Chinese wampee and White sugar Chinese wampee average pollen tube length in comparison with that of Seedless Chinese wampee was 2. 0 times. The pollen activity of Chinese wampee which determined by different dyeing method exist difference in a certain degree.
