抗碱枳砧突变体的RAPD分析及其特异片段克隆

【摘要】 利用10个随机引物对枳砧,酸橘及抗碱枳砧突变体基因组DNA的多态性进行RAPD分析。其中2个引物能够检测出三者间DNA的多态性,分离出1.8~2.2 kb和0.6~1.1 kb的特异DNA片段。将800 bp特异DNA片段克隆到pUC19载体,并作了主要酶切位点分析。
【Abstract】 RAPD markers were generated for identifying Poncirus trifoliate mutant resistant to akali. Almost all 10 single random primers tested exhibited the same and similar amplified DNA patterns except that two primers were screened to distinguish three cultivars, the 1. 8-2. 2 kb and 0. 6-1.1 kb DNA specific fragments were separated. The 800 bp specific fragments was cloned into pUCl9 vector and analysized with restriction endounclease.
