黄瓜全雌性基因连锁的AFLP和SCAR分子标记

【摘要】 黄瓜是重要的蔬菜作物,性型表现多样。黄瓜全雌性品系在简化杂交制种、提高产量等方面均具有重要的作用。与全雌性连锁的分子标记辅助育种将加速全雌品系的获得。本研究以来源于全雌EC1和弱雌CC3杂交的F2性型分离群体为材料,利用BSA法,筛选了64对AFLP选择性引物EcoRI-NN+MseI-NNN组合,发现EcoRI-TG+MseI-CAC引物组合在全雌基因池中扩增出一条分子量为234bp的特异带,而在雌雄同株基因池中未见。经F2代性型分离群体单株验证,MAPMARKER(Version 3.0)软件分析,该标记与全雌性位点的连锁距离在6.7cM。命名该连锁标记为TG/CAC234。将该特异条带回收、克隆、测序,设计特异SCAR引物,再对F2代单株基因组DNA进行扩增,仅在全雌单株中扩增出一条特异带。因此,成功地将与黄瓜全雌性连锁的AFLP标记转化为操作简便、表现稳定的SCAR标记。该SCAR标记被命名为SA166。

【Abstract】 In this study the amplified fragment length polymorphism (AFLP) technique with 64 primer combinations and bulked segregant analysis were employed to find the polymorphisms between the DNA pools of gyneocious and moneocious cucumber from F2 population derived from a cross of CC3 × EC1. The primer combination of TG+CAC could stably amplify a specific fragment, TG/CAC234 in gyneocious DNA pool. The TG/CAC234 marker was testified to be closely linked to the gyneocious loci with genetic distance of 6.7 cM through the analysis of the F2 population using the software of MAPMARKER (version 3.0). TG/CAC was sequenced and the sequence information was used to design sequence-characterized amplified region (SCAR) primers. Amplification of different sex phenotypes of F2 population proved the SCAR marker, named SA166, is specific for gyneocious gene. This marker has been used successfully in marker-assisted selection in the cucumber breeding program.
