番茄几丁质酶基因转化西瓜研究初报
A preliminary study on genetic transformation of watermelon with tomato chitinase gene

【摘要】 本试验构建了以CaMV35s启动子驱动的含有番茄酸式胞外几丁质酶基因(CHI3)的植物表达载体,通过冻融法导入根癌农杆菌(Agrobacterium tumefaciens)菌株EHA105中,用叶盘法转化西瓜(Citrullus lanatus(Thunb.)Mansfeld var.citroides)栽培种“中育1号”,通过卡那霉素抗性鉴定、CHI3基因的PCR检测、Southern斑点杂交、RT-PCR检测证实CHI3基因已经整合到西瓜基因组中,并获得表达。
【Abstract】 The expression vector p121CHI3 carried tomato acidic intracellular chitinase gene(CHI3) under the drive of CaMV35s promoter, was successfully transferred into watermelon cultivars "ZhongYuYiHao" via Agrobacterium-mediated transformation. Transgenic watermelon plants were confirmed by the Kanamycin-resistant experiment, PCR analysis , Dot blotting and RT-PCR.
