通过分生组织培养脱除(艹晶)头(Allium chinense G.Don)病毒

Elimination of Viruses from Infected Bulbs of Allium chinense G.Don by in vitro Shoot Apex Culture

【摘要】 藠头茎尖(含l~2个叶原基)在B5,N6,MS和农事培养基上均能生长,但B5培养基最适宜,农事培养基最差。所得茎尖再生株在1/2MS无激素培养基上迅速生根,移栽后100%存活。由于藠头行无性繁殖,日间调查已普遍受病毒病感染,病毒粒体线状,局部侵染苋色藜、昆诺阿藜和千日红。以其为指示植物对茎尖培养再生株进行检测,无病症植株比率为61%。

【Abstract】 Shoot apexes (with 1 - 2 leaf primordium) of Allium chinense G.Don were excised and cultured on different medium for plant regeneration. Results showed that growth of the explants on B5 medium is best but worst on Nongshi medium and middle on MS and N6 medium.Regenerated plants rooted directly in 1/2 MS medium with no hormones.The recovering rate reached to 100% when these plants transplanted to soil.Electron microscope observation revealed that virus infecting Allium chinense was a kind of linear particles,and infecting Chonopodium amaraanticoloo,Chonopodium quinoa.and Clobeamaranth easily.Using them as the indicating plants, 61% of them showed no symptom when different leaf extraction samples from the regenerated Allium chinense plants were inoculated on their leaves.
