地灵试管克隆繁育研究

【摘要】 研究证明,地灵地下根状茎用75%乙醇和0.1%升汞分别消毒25~30s和16~18min,可获得较好的消毒效果,并且经适当激素诱导即可建立起离体克隆繁育体系;6-BA与KT配合使用,能诱导地灵茎及组织产生大量不定芽,但对不定芽的进一步生长、发育不利,1.0~1.5mg·L-1KT与0.5mg·L-1IAA配合,可诱导地灵不定芽增殖并形成发育健壮的试管苗,增殖效率达4.20~4.67,苗高2.25~2.84cm。

【Abstract】 It was testified in this research that the rhizoma of stachys disinfected respectively for 25 - 30 seconds and 16 - 18 minutes by 75% alcohol and 0.1% mercuric chloride could get better disinfectant effect.The system of In vitro propagation could be set up through proper inducement from hormone. Assortative usage of 6-BA (6-benzylaminopurine) and KT (6-Furfury-laminop-urine) could cause rhizoma of stachys to produce lots of indefinite bud, but it was disadvantageous for father differentiation, growth and developing of the indefinite bud.The concentration of KT with 1.0 - 1.5mg· L-1 and IAA (3-Indolebutyric acid) with 0.5 mg· L-1 could induce indefinite bud of stachys to proliferate and to develop into healthy seedling.The generation rate was 4.20 -4.67 and seedling height was 2.25 - 2.84 centimeter.
