稀有香辛植物山葵(Wasabi japonica Matsum)的茎尖离体培养研究

【摘要】 利用山葵顶芽及侧芽生长点诱导山葵无菌苗,对无菌苗进行不同激素种类和浓度试验,结果表明,BA,KT,ZY三种细胞分裂素中,BA和KT对山葵增殖效果较好,NAA,IAA,IBA三种生长素中,以NAA和IBA对增殖效果较好,最佳增殖培养基为MS+BA 0.5 mg·L-1+NAA 0.05 mg·L-1;MS+KT0.5 mg·L-1+NAA 0.05 mg·L-1;CPPU对山葵增殖有促进作用,最适宜培养基为MS+CPPU 0.4mg·L-1+NAA 0.05mg·L-1;BA与KT结合使用对增殖效有利,MS+BA 0.2mg·L-1+KT0.5 mg·L-1+NAA 0.05mg·L-1的激素组合繁殖系数可达5.1;增殖培养以培养基pH5.8,光照1500Lx,18℃培养最适宜增殖;添加0.02mg·L-1的GA3可以促进山葵试管苗的生长。生根试验表明,较低浓度的生长素对生根有利,最佳生根培养基为MS+NAA 0.05 mg·L-1或IAA 0.05 mg·L-1或IBA 0.05mg·L-1,生根率可达100%,试管苗进行驯化移栽,成活率达90%。
【Abstract】 By using apex buds and lateral buds of wasabi as explants,the micropropagation indice were improved from 3.0 - 4.0 to 4.0-5.0 with auxin, cytokinins and new plant regulators by new methods.In order to improve the rates, some experiments by using different hormones and concentrations were done.The results showed that BA and KT were best among BA, KT,ZT, NAA and IBA were so among NAA,IAA and IBA.The better media for plantlets propagation were MS+BA 0.5mg·L-1 + NAA0.05 mg· L-1 or MS + KT0.5 mg· L-1 + NAA0.05 mg· L-1; CPPU can enhance the Wasabi propagation and the suitable medium was MS + CPPU0.4 mg·L-1; BA combined KT were better, and the propagation rates on MS + BA0.2 mg·L-1 + KT0.5 mg·L-1 + NAA0.05 mg·L-1 could reach 5.1.From the experiments,it was suggested that pH5.8,light 1500 lux,temperature 18℃ were best for plantlet propagation.The experiments result about root growth showed that auxins at low concentration was helpful and the better medium was MS+ NAA0.05 mg·L-1 or MS+ IAA0.05 mg·L-1 or MS+ IBA0.05 mg·L-1.On these media,the root growth rates can reach 100% and the living rates can reach 90% when the plantlets were trained and planted.
