非洲菊的组织培养和快繁技术
Effect of Different Media Using in Gerbera Tissue Culture and Production

【摘要】 试验以非洲菊幼嫩花托为外植体,以MS为基本培养基,添加含不同浓度的6-BA,KT,IAA,NAA,IBA等激素诱导成芽,进而继代扩繁。比较了不同生长激素及其含量对诱导分化生芽、生根的效果和对移栽成活率的影响。采用二次灭菌方法,灭菌成功率提高35.5个百分点;采用MS+6-BA 10+IAA0.5诱导培养基,试验的六个品种均诱导成芽,不同品种的诱导率表现出差异,愈伤组织诱导率在50%~100%之间;在芽的生长和增殖阶段采用BA0.5~1.0+IAA0.1的组合有较高的增殖倍数且苗在生长速度、叶型、植株叶色、脆性等方面均有较好的表现,在相同的培养基条件下,各品种间芽的增殖和生长存在着差异;生根培养基以MS添加较低浓度的IAA0.5或IBA0.1mg/L生根快、根粗细适中,移栽成活率高。
【Abstract】 The effect of cytotins and auxins using in rapid propagation of Gerbera jamesonii Bolus tissue culture was studied.MS media supplemented with 6-BA 10mg/L + IAA 0.5mg/L in order to induce callus for producing the shoots through young capitulum explants. In gerbera proliferation media the effect of combining with 6-BA 0.5 - 1.0mg/L + IAA 0.1 mg/L was better than that of only using higher concentration of 6-BA 2.0mg/L or combining with KT 5 - 10mg/L + IAA 0.5mg/L.The result shows that low concentration of IAA 0.5mg/L or IBA 0.1mg/L can promote to grow roots, result in 100% rooting rate and 100% survival rate of transplanted plantlets.The six varieties show difference during the propagation in the same medium in vitro.
