蕙兰×台兰种间杂交种子无菌播种育苗技术研究
A study on the aseptic Seeding of interspecific hybridization seed from "Huilan" (Cymbidium faberi) and "Tailan" ( C. floribundum var.pumilum) orchids

【摘要】 以蕙兰x台兰种间杂交的种子为材料,研究适合于种子无菌播种、原球茎分化芽、生根壮苗的不同培养基.在5种用于无菌播种的培养基中,添加IAA10mg/L的C1基,种子萌动率达3%,原球茎由培养120天的10.13个/瓶,平均直径1.63mm到220天增至19.86个/瓶,平均直径4.30mm;BA1~2mg/L有利于原球茎分化芽,分化率达100%以上;NAA2mg/L使生根壮苗率达100%.添加活性炭1.5~3g/种子成苗效果较好,在芽分化与生根壮苗培养中,减少MS大量元素的使用量有利于生长.兰花试管苗移栽成活率为80%,已获得4~5年生的杂种苗.
【Abstract】 Various media were tested tor their suitability in aseptic seeding, bud ditterentia-tion of protocorm and root induction culture with interspecific hybridization seed from "HuiLan"and"Tailan"orchids.The results showed that among five kinds of media Cl(10mg/ LIAA) for aseptic seeding was better than others, the rate of protocorm forming from seed being as high as 3% , the number of protocorm increasing from 10.13 per plate at 120 days to 19.86 per plate at 220 days after culture, and at the same time, the average diameter of protocorms increased from 1.63mm to 4. 3mm; while 1 ~ 2mg/LBA was suitable for bud differentiation of protocorms with bud differentiation rate;2mg/LNAA was suitable for root induction. Favorable effect of active carbon (1.5~3g/L) on hybrid seedling culture was observed . Decrease in usage of macro - elements favors bud differentiation, root induction, and growth of seedlings as well. The transplanting survival rate of these seedlings was 80%. These seedling have grown for more than five years.
