草莓花药愈伤组织细胞悬浮系的建立

【摘要】 以草毒品种宝交早生为试材,用F培养基,附加BA2.0mg/L,NAA0.2mg/L,CH300mg/L进行花药愈伤组织诱导,其出愈率达89.13%.以MS为基本培养基,通过改变培养基中的激素种类及浓度、有机附加物的种类及浓度、无机盐的种类和浓度,愈伤组织在继代培养过程中的状态不断变化,并诱导出胚性愈伤组织.胚性愈伤组织在MS无机盐+B5有机物+2,4-D1.0mg/L+NAA1.0mg/L+ZT0.2mg/L+CH400mg/L+Gln150mg/L+KCl400mg/L+蔗糖3%(PH=5.8)的液体培养系中培养一个月,建立了细胞悬浮系,并诱导发生了胚状体.

【Abstract】 Callus formation of strawberry ( Fragaria ananassa Duch, cv: Houkouwase) was achieved using F medium added with BA2. 0mg/l, NAA0. 2mg/l, CH300mg/. The callus induction rate was 89.13% , The state of the callus was regulated by modificating the con-centration of hormones, organic and inorganic additives during subculture and embryogenic callus was gained from Houkouwase anther callus. Cell suspension was established by cul-turing embryogenic callus in liquid medium containing MS salts + B5 organic additives + 2, 4 - D1. 0 mg/L + NAA1. 0 mg/L + ZT0. 2 mg/L + CH400 mg/L + Gln150 mg/L + KCL400 mg/L + surose3 % , pH = 5.8, Embriyoids were formed from thesuspension during culturing
