外源DNA导入在葡萄抗病育种上的应用研究
Studies on the Techniques of Introducing Ex-ogenous DNA Applied in Grape Resistance Breeding

【摘要】 以藤稔葡萄为DNA导入的受体,以毛葡萄为DNA导入的供体,以藤稔葡萄单芽茎段为外植体,在组织培养中进行外源DNA的导入.研究DNA溶液不同浓度与时间处理对腋芽萌发和生长的影响,并对处理进行POX、SOD两种酶活性与同工酶分析,以检测其抗病性与变异情况.结果表明:①DNA溶液对腋芽的萌发有明显的抑制作用,且随DNA浓度的升高和处理时间的延长萌发率呈下降的趋势,但以DNA400μg/L浸渍处理3~5d腋芽既能保持一定的萌发率,表型也发生一定的变化;②导入材料POX、SOD活性明显高于对照,同工酶谱也存在着明显差异,发DNA 400μg/l浸渍腋芽5d,POX、SOD活性入及同工酶谱与对照差异明显.
【Abstract】 This experiment has following contents: Using improved Variety Fujiminori, as the DNA receptor and V.quingquangularis Rehder (with high resistance to Anthracnose) as the DNA donor, Using stem segment with axillary bud as explant, adopting penetrating growing - point method to introduce exogenous DNA in the course of tissue culture. Studing the influence of different DNA concentrations and different treated time on germination and growth of axillary bud, analysising the activity and isoenzyme of Peroxidase and Superoxidese of the treated materials in order to check its resistance and variation. The main results were obtained as follows:( 1 ) Exogenous DNA reduces obviously the germination of axillary bud and the germi-nation rate will reduce with increasing DNA concentration and prolonging the time of DNA treated. But the axillary hud can keep a certain germination rate and the phenotype will change in certain extent with DNA concentration 400ug/l penetrating 3 - 5d.(2) There is difference of activity and isoenzyme bands of Peroxidase and Superoxidase between the materials introduced exogenous DNA and the check. And there is obvious dif-ference between DNA 400ug/l penetrating axillary bud 5d and the check.
