安梨不同器官、不同发育时期的过氧化物酶同工酶

【摘要】 本试验测定了安梨不同器官、不同时期的过氧化物酶同工酶谱。结果表明,安梨各器官组织的过氧化物酶同工酶共表达12条酶带,依其Rf值可大致分为三区;不同器官组织中既有共同酶带也有一定的差异,其中以皮层、叶片的同工酶表达最多,分别为8条和9条;以果心、芽和木质部中表达最少,分别为1-3、4和3条酶带。各器官组织中共有的酶带即安梨特征酶带为9、 10和11号酶带。不同生育期各器官组织的同工酶谱有一定的变化,叶片中基叶酶带较多,中部叶次之,嫩叶最少;果实发育过程中酶带数呈减少的趋势;夏芽酶带较少,而秋季的芽中酶带效目明显增多。总的规律是幼嫩、生长迅速的器官组织酶带少、变异大,而成熟尤其是停止生长的器官组织酶带较多并趋于稳定。另外,试验还证明安梨的过氧化物酶同工酶在一定程度上受环境因素的影响,所以在进行无性系鉴定时,应注意样品所处环境的一致性。重复性试验表明,同一发育时期的同一器官组织的酶谱表达基本一致,说明过氧化物酶同工酶确实反映了物种本身的遗传特性。

【Abstract】 In the test the peroxidase isoenzymes of different organs at the different development periods of Anli pear (Pyrus nssuriensis Maxim) were determined. The results suggested that the organs expressed altogether 12 bands. According to the Rf values of each band, they could be divided into three regions. The cortex and leaf had the most isoenzymes ; fruit kernel, budand xylem expressed fewest. At different development periods the isoenzymes of different organs changed to a certain degree. The general rule was that the number of the bands in the young tissues that grew rapidly were less and varied greatly, in the mature tissues were more and trended to steady. In addition, it was proved that the expression of isoenzymes was affected to a certain extent by the environment, so that the expriment conditions must be kept consistent when the strains or clones are identified.
