叶酸、维生素B6、维生素B12体外对同型半胱氨酸致大鼠主动脉内皮细胞损伤的保护作用

【摘要】 目的观察叶酸(FA)、维生素 B6(VB6)、维生素 B12(VB12)对同型半胱氨酸致血管内皮细胞损伤的交互保护作用。方法原代培养大鼠主动脉内皮细胞,MTT 法观察叶酸、VB6、VB12各自单独5 个剂量(加上阴性阳性对照组,共7组)对 Hcy 抑制大鼠主动脉内皮细胞(RAEC)增殖的保护作用。然后采用 L27(313)正交设计,通过 MTT 细胞增殖实验观察叶酸、VB6、VB12的交互作用;根据正交分析结果分为6组,三组维生素防护组、阴性对照组、阳性对照组及维生素对照组,维生素用含终浓度为1.0mmol/ LHcy 的培养基配制,干预24h 后,取上清液,比较各组超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶 (GSH-Px)、丙二醛(MDA)、一氧化氮(NO)、一氧化氮合酶(NOS)等指标的差异。结果叶酸与 VB12能够缓解1.0mmol/L Hcy 对大鼠主动脉内皮细胞(RAEC)增殖的抑制作用,而 VB6则无明显效果;叶酸与 VB6 无交互作用,与 VB12存在交互作用;VB6与 VB12之间也存在交互作用;FA、FA+VB12、FA+VB6+VB12三种组合均能拮抗 Hcy 对 SOD、GSH-Px、NOS 酶活性的抑制作用,保护内皮细胞功能。结论同时补充叶酸、 VB6、VB12可能是三者在体外保护 Hcy 损伤内皮的较理想组合。
【Abstract】 Objective to investigate the cross protective effects of folic acid,VB6,VB12 on rat aortic endothelial cells injuried by DL-homocysteine (Hcy).Method Rat aortic endothelial cells (BAEC) were cultured in vitro,and the cross effects of folic acid,VB6,VB12 were studied according to orthogonal design by MTT. According to the results of orthogonal design,the activities of SOD、GSH-Px、NOS and the contents of MDA、NO in the medium were compared.Results The results of orthogonal design indicate that,the inhibition of DL- homocystine to RAEC could be relieved by folic acid,VB12 excluding VB6.The interaction existed between folic acid and VB12 as well as VB6 and VB12.The inhibition of Hcy to the enzymes (SOD、GSH-Px、NOS) could be relieved by three vitamin projects.Folic acid,VB12,VB6 could inhibit the reduction of NO and decrease the production of MDA.Conclusion The simultaneous addition of folic acid,VB6 and VB12 may be an excellent strategy for protection of RAEC injuried by DL-homocysteine.
