茶多酚促排镉作用的实验研究

【摘要】 目的探讨茶多酚促排镉作用及其对肝肾损伤的影响。方法 60只清洁级SD系雄性大鼠,按体重大小随机分为6组:(Ⅰ)正常对照组,(Ⅱ)茶多酚组,(Ⅲ)单染镉组,(Ⅳ)低剂量荼多酚干预组, (Ⅴ)中剂量茶多酚干预组,(Ⅵ)高剂量荼多酚干预组。Ⅲ-Ⅵ组以CdCl2腹部皮下注射染毒5周,染毒的同时分别予低、中、高剂量茶多酚灌胃,Ⅱ组给予中剂量茶多酚灌胃,Ⅰ、Ⅲ灌以等量的蒸馏水。干预结束后,收集24h尿液、粪便、并处死大鼠取肝、肾、睾丸等样品进行相关指标的测定。结果 (1)中高剂量干预组肝镉和睾丸镉含量低于单染镉组下降(P<0.05),尿镉和粪镉排出量高于单染镉组(P<0.05)且具有量效关系。而肾镉、骨镉含量各干预组与单染镉组之间未见显著性差异(p>0.05);茶多酚组肝镉含量尚低于正常对照组(P<0.05)。(2)肝肾损伤检查:血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、血清尿素氮(BuN)、肌酐(CRE)干预组与单染镉组之间未见显著性差异(p>0.05)。结论中高剂量茶多酚可促进染镉大鼠体内镉排出及正常大鼠体内镉排出趋势,具有较强驱Cd作用。且驱Cd同时不加重肝肾损伤。其机理可能与茶多酚螯合金属离子的作用有关。
【Abstract】 Objective To study the effects of tea polyphenols (TP) on promoting excretion of cadmium and its influence on the liver and kidney. Methods 60 male Sprague-Dawley rats were randomly divided into six groups; control, TP, Cd, low-dose TP intervenient, middle-dose TP intervenient, high-dose TP intervenient groups. The rats in control and TP groups were injected (sc) with saline, while others with Cd at a dose of 0. 4mg/kg,. . Mean while, the rats inlow-dose TP intervenient, middle-dose TP intervenient , high-dose TP intervenient and TP groups were given TP ( 50 mg/kg, 100 mg/kg, 200 mg/kg ,100mg/kg respectively) orally once a day , while others were given distilled water instead. After five weeks , the rats were killed and the samples of serum, 24h urine, feces, liver, kidney and testis were taken for test. Results (1) The Cd concentration of liver and testis in middle-dose and high-dose TP intervenient groups were lower than that of Cd group (P < 0. 05). Mean while , The Cd concentration of urine and feces in these two groups were higher than that of Cd group (P < 0. 05), but there were no statistical difference among all group as for kidney Cd concenration. (2) the serum level of ALT、 AST、 BUN、 CRE were no diffence among all groups. Conclusion TP could promote accumulative cadmium to excretion without increasing the hurt of liver and kidney. The mechanism maybe related to its chelating effect.
