软骨藻酸诱导PC12细胞凋亡作用及对细胞周期的影响

【摘要】 目的:研究软骨藻酸诱导PC12细胞凋亡作用及对细胞周期和活性氧生成的影响。方法:流式细胞仪法测定0、0.01、0.1、1μmol/L软骨藻酸作用细胞24h后对细胞凋亡及细胞周期和活性氧生成的影响。结果:软骨藻酸可诱导PC12细胞凋亡的发生,所有剂量组的凋亡发生率与对照组比较,差异具有显著性。流式细胞仪法测定软骨藻酸对PC12细胞细胞周期的影响:软骨藻酸可将PC12细胞阻滞于细胞周期中的S期,中、高剂量组中处于S期的PC12细胞数量和百分比与对照组比较,差异具有显著性。软骨藻酸对PC12细胞活性氧生成的影响:软骨藻酸对PC12细胞ROS的产生具有一定的影响,高、中、低剂量组均可使PC12细胞的ROS产生量增加,与对照组相比均具有显著意义。结论:软骨藻酸能诱导PC12细胞凋亡的发生,将PC12细胞阻滞于细胞周期中的S期,并使得细胞内的ROS产生增加。
【Abstract】 Objective: To study the apoptotic PC12 cells induced by Domoic acid and the effect of Domoic acid on the cell cycle and reactive oxygen species in PC12 cells. Methods: The apoptotic PC12 cells, the cell cycle and reactive oxygen species was measured by the Flow cytometric analysis treated with 0、0.01、0.1、1μmol/L Domoic acid 24h. Results: Flow cytometric analysis indicated that the Domoic acid could induce apoptosis of PC12 cells. The effect on the cell cycle: The cell cycle was blocked markedly at S phase after exposing PC12 cells to Domoic acid for 24 h specifically in the middle and high concentrations. Reactive oxygen species (ROS) induced by Domoic acid: Domoic acid had an effect on ROS in PC12 cells, and three dosages exhibited more elevated ROS level than that of the control group. Conclusion: Domoic acid could induce apoptosis of PC12 cells, block the cell cycle at S phase and elevate ROS the level in PC12 cells.
