二氧化硅对人呼吸道上皮细胞及巨噬细胞内IL-8和TNFα的诱导作用

【摘要】 目的:探讨二氧化硅对人支气管上皮细胞(BEAS-2B)、肺泡上皮细胞(A549)及巨噬细胞(THP-1)内白细胞介素-8(IL-8)与肿瘤坏死因子(TNFα)基因表达的诱导作用。方法:以人呼吸道上皮细胞及巨噬细胞作为靶细胞,应用定量PCR技术检测二氧化硅对细胞内IL-8及TNFαmRNA表达的诱导作用。结果:二氧化硅可提高IL-8及TNFαmRNA在三种受试细胞内的表达水平。以相应对照组mRNA水平为参照,在同一试验条件下,三种细胞在二氧化硅刺激后细胞内IL-8及TNFαmRNA相对表达强度不同,表现为:THP-1>BEAS-2B>A549。结论:二氧化硅可显著提高矽肺发病相关细胞因子IL-8及TNFα基因的表达水平。二氧化硅的上述作用,以对巨噬细胞的诱导作用为最强。
【Abstract】 Objective: To explore the effect of silica on the expression of interleukin 8 (IL-8) and tumor necrosis factor (TNFα) genes in human bronchial and alveolar epithelial cells, and macrophages. Methods: Human bronchial epithelial cells (BEAS-2B), alveolar type II epithelial cells (A549), and macrophages (THP-1) were stimulated with silica. Total RNA was extracted and mRNA transcribed to cDNA. Levels of mRNA were determined using quantitative PCR. Result: Silica exposure significantly increased levels of IL-8 and TNFα mRNA in three cell types. However, the potency that silica presented in induction of IL-8 and TNFα gene expression appeared to be stronger in macrophages than in bronchial and alveolar epithelial cells. Conclusion: Silica dusts induce differential expression of pneumoconiosis-related cytokines including IL-8 and TNFα in different airway epithelial cells and macrophages. This study provides information used for prevention and treatment of pneumoconiosis.
