通过基因芯片筛选获得与PmLK906连锁的ESTs

【摘要】 普通小麦(Triticum aestivum L.)品系‘兰考906’携带一个小种专化的隐性抗白粉病新基因,暂称为 PmLKg06.用‘中国春’ב兰考906’F2:3纯合抗、感家系构建抗、感 cDNA 池.以此抗、感 cDNA 池为探针筛选小麦基因芯片,获得了可能与 PmLK906基因连锁的 EST(expression sequence tag)信息.其中感病池表达量高8倍以上的34个,抗病池表达量高8倍以上的28个,排除假阳性结果,这些信息可能代表了与 PmLK906紧密连锁的基因.从抗病池表达量高的28个 EST 中选择2个 EST 序列信息,设计了2对特异性引物,在抗、感亲本 cDNA 和抗、感 cDNA 池中扩增,显示出一致的差异条带,提示与 PmLK906连锁.

【Abstract】 Common wheat(Triticum aestivum L.)lines of’Lankao 90(6)’harbor a novel recessive powdery mildew resistance gene temporarily named PmLK906.Powdery mildew susceptible cDNA pool and resistant cDNA pool were established using homozygote susceptible hnes and resistant lines derived from a cross of’Chinese Spring’×’Lankao 90 (6)21-12’.The susceptible and resistant cDNA pools were used as probes to screen the Affymetrix wheat gene chip. 34 ESTs of 8 times or more highly expressed in susceptible cDNA pool,and 28 ESTs in resistant pool were obtained. Except fail positive ESTs,the others maybe representatives of the genes closely linked to PmLK906.Two specific primer pairs were designed basing on two ESTs selected from the 28 highly expressed in resistant cDNA pool.The amplification results,using cDNAs of susceptible/resistant parents and pools as templates,showed coincident polymorphic bands,indicating they were linked with PmLK906.
