分子标记在优质蛋白玉米(QPM)育种中的应用

【摘要】 利用CIMMYT开发的opaque-2(o2)SSR标记,可以有效地用于优质蛋白玉米育种。本研究利用SSR标记,检測了QPM群体中群13、中群14内o2基因的频率,发现中群13、中群14发生了o2基因的遗传漂变。检测QPM高代系CA、CD、CB、R中的o2基因频率,发现在CA、CD、CB、R高代系中o2o2基因型频率分别为91.7%、80.0%、0.0%和100%,表明用常规方法选育QPM自交系,不能保证纯合o2基因的频率。为了拓宽QPM种质基础,将普通自交系转化为QPM自交系,利用分子标记追踪普通玉米与QPM自交系回交后代的o2基因,结果表明是非常有效的。
【Abstract】 The SSR markers developed by CIMMYT can be effective used in the QPM breeding efforts. In the study, the SSR markers was used in the evaluation of the inbred lines developed in recent years, and QPM breeding program to change the normal elite lines (NELs) into QPM lines. After detected frequency of o2 gene in QPM populations CPop13 and CPop14, and inbred lines CA, CD, CB, R, genetic drift was found, and the o2 genotype frequency in CA, CD, CB, R was detected as 91.7%, 80.0%, 0.0%, 100% respectively. It was proved thai homozygous o2o2 genotype in QPM could not be ensured lines in conventional breeding. In order to extend QPM germplasm and transform normal elite lines into QPM lines, the markers was used in the detection of o2 gene in backcross generations of conventional lines and QPM lines, and it was proved a powerful tool in the effort.
