丹参等几种中药指纹图谱与代谢指纹图谱研究

Fingerprint and metabolic fingerprint analysis of Danshen root and several other traditional Chinese medicines by HPLC-UV and HPLC-MS

【摘要】 应用高效液相色谱技术对丹参、野生人参、三七、银杏叶等药材和相关制剂进行了指纹图谱和代谢指纹图谱的研究。对不同产地的丹参野生和种植的药材样品进行了详细的分析比较研究,发现不同产地的药材在指纹图谱上存在较大差异,还发现其中多酚酸类成分在植物中的存在状态不同。不同生产厂家生产的丹参注射液和香丹注射液的指纹图谱差异较大,研究结果表明生产工艺的不同是造成产品指纹图谱差异的主要原因,对成品指纹图谱的影响比因产地不同造成的影响大。应用高效液相色谱-质谱联用技术对指纹图谱中的主要色谱峰进行了指认。另外还对丹参注射液给大鼠后在体内的分布和代谢指纹图谱进行了研究,发现注射给药后丹参中的主要成分丹酚酸B迅速分解代谢成其他化合物,而丹酚酸A保持相对较高的浓度。此外还应用高效液相色谱技术对野生人参、三七、银杏叶、降香、芍药、川芎等药材的指纹图谱进行了研究。上述研究对于有效控制这些中药及其相关制剂的质量奠定了科学基础。

【Abstract】 HPLC-UV and HPLC-MS techniques were used for fingerprint and/or metabolic fingerprint analysis of Danshen root(Salvia miltiorrhiza) and its injection,wild ginseng,notoginseng,Gingko biloba leaf,rhizome of Lingusticum chuanxiong and lignum of Dalbergia odorifera.HPLC profiles of Danshen injections from a pharmaceutical company and from several different companies were established and employed to analyze their consistency.The results showed that Danshen injections from different companies possessed very different HPLC profiles but from the same company with different batches had similar HPLC fingerprint.Eleven common peaks were recognized and identified by HPLC-MS analysis and by comparison with reference standards.The crude drug of Danshen was also analyzed under the same HPLC conditions.The results showed that the chemical constituents in both Danshen injection and the crude drug were well separated and detected by the established HPLC-UV and HPLC-MS methods.The main constituents are different between Danshen injection and the crude drug.This indicated that the great changes of main chemical constituents had taken place in the course of preparation.It is helpful to optimize the fingerprint model to apply both HPLC-UV and HPLC-MS technology for fingerprint analysis of Danshen injection.The established HPLC-UV fingerprint analysis could be successfully employed to evaluate and control the quality of Danshen injection and its crude drug.In order to obtain more information about the active constituents of Danshen injection in biological system, the rat’s plasma was analyzed by HPLC-UV and HPLC-MS after intravenous administration of Danshen injection at different time interval.Except for protocatechuic aldehyde,other major chemical constituents in Danshen injection appeared in rat’s plasma after intravenous administration,but the quantity of each main constituent was greatly different from Danshen injection.With the administration time prolonged,salvianolic acid B was metabolized at fast rate. While salvianolic acid D,lithospermic acid and salvianolic acid A eliminated at slow rate and still maintained relatively high quantity after 30-minute intravenous administration.Fingerprint technique was also employed to analyze the samples of Panax notoginseng root,wild Ginseng, Gingko leaf,rhizome ofLingusticum chuanxiong and lignum ofDalbergia odorifera.
