关于HCV不同区段抗原的反应性研究

【摘要】 目的探讨各种HCV抗原在检测中的作用及其与HCV RNA的关系。方法利用HCV不同区段抗原抗-HCV酶联免疫试验(EIA)和逆转录聚合酶链反应法(RT-PCR),对不同临床型HCV感染者进行检测。结果各组病人均以抗-NS3和抗-C阳性率最高。但均有一定比例病人抗-NS4和抗-NS5阳性。与各单个片段抗-HCV阳性率相比,抗-HCV总抗体阳性率最高。结论NS3和C抗原在测定抗-HCV中起很重要作用,但NS4和NS5抗原在抗-HCV EIA中的作用也不容忽视。联合应用不同区段HCV抗原将大大提高抗-HCV EIA的灵敏度。
【Abstract】 Objective Study the roles of different HCV antigens and their relationship with HCV RNA.Methods Serum samples from patients with acute,chronic hepatitis,HCV related hepatocellular carcinoma and HCV carriers were tested for antibodies against antigens encoded by HCV different gene fragments using enzyme immunoassays(EIA),and for HCV RNA by a reverse trascription polymerase chain reaction(RT-PCR).Results The positive rates of anti-NS3 and anti-C were the highest in four groups of patients as compared with anti-E1,E2,NS4 and NS5.A proportion of patients in the four groups was anti-NS4 and/or anti-NS5 positive,although other HCV antibodies were not detected.The positive rate of total anti-HCV was much higher than that of antibodis against any one of antigens encoded by HCV gene fragments.Conclusion NS3 and C antigens are more important for detection of anti-HCV.However,the role of NS4 and NS5 antigens should not be neglected.Combination use of different HCV antigens in anti-HCV EIA greatly increases the sensitivity of the kit.
