体外彗星试验检测34种化合物对小鼠淋巴细胞的遗传毒性

【Abstract】 The single cell microgel electrophoresis assay(comet assay) can detect DNA single strand breaks,alkaline labile sites and other genetic damages.Compared with traditional DNA break test methods,comet assay is more quick,sensitive,convenient and visual.Particularly,it is applicable to various mammalian cell types,needs much less cells,and does not need radio-active isotope labeling.It can study the genotoxic specificity at the single cell level,therefore can be used to screen drug genotoxity.In this study,the genotoxicities of 34 chemicals to mouse lymphocytes were further tested by in vitro comet assay.The test substance included known genotoxicants, antineoplastic drugs,and flavone derivatives.Among the test chemicals,the concordance of present study with Ames test was 62.1%(18/29),and the comparison of present results with animal carcinogenecity test data showed that the sensitivity was 75.0%(15/20),and the concordance,73.9 (17/23),respectively.Flavone derivatives SIPI-644(10μM) and SIPI-1211(33μM ) were positive in the presence of S9 while others were negative.Therefore the comet assay can be used to detect genotoxicants of various mechanisms including free radicals,alkylating of nucleotide,DNA adduct, strand break,etc.This indicated that further in vivo model could be explored to screen drug genotoxicity when combined with the general toxicity test and the pharmacokinetic study.
