鸡ACOX3基因5′侧翼区多态性与生长和体组成性状的相关研究
A Single Nucleotide Polymorphism of Chicken Acyl-Coenzyme A Oxidases 3 Gene 5’ Flanking-Region Associated with Growth and Body Composition Traits

【摘要】 本研究旨在分析酰基辅酶A氧化酶(acyl-Coenzyme Aoxidases 3,ACOX3)基因5′侧翼区单核苷酸多态性与生长和体组成性状的关系,以期了解该基因与脂类代谢的关系。本研究以东北农业大学肉鸡高低脂双向选择品系和东北农业大学F2资源群体为材料,采用PCR-RFLP方法检测了ACOX3基因5′侧翼区的多态性,并分析基因多态性与鸡生长和体组成性状的相关。结果发现一个点突变c.-442A>C。该突变位点在高、低脂双向选择品系八、九、十世代混合群体对5周龄和7周龄体重、屠体重、小胸肌重、小胸肌率、脾脏重、脾脏比率有显著影响(P<0.05);对1周龄和3周龄体重、大胸肌重、大胸肌率、腿肌率、心脏重、心脏比率有一定影响(P<0.2)。该突变位点在F2资源群体对初生重、10-12周龄体重、屠体重、小胸肌重、小胸肌率、4周龄跖骨围、6周龄跖骨长、10周龄跖骨长、12周龄跖骨长、股骨长、胫骨长、胫骨重有显著影响(P<0.05);对8周龄体重、9周龄体重、8周龄跖骨长、8周龄跖骨围、跖爪重、跖爪率有一定影响(P<0.2)。ACOX3基因对鸡生长和体组成性状具有广泛的影响;该基因的c.-442A>C突变尤其对鸡体重和肌肉重有显著的影响,可以尝试作为遗传标记对体重和肌肉重性状进行分子标记辅助选择。
【Abstract】 This experiment was conducted to study the associations of polymorphism of Acyl-Coenzyme A oxidases 3(ACOX3)gene 5’ flanking-region with growth and body composition traits in chicken,in order to understand the relation between this gene and lipid metabolism.In the present study,the Northeast Agricultural University broiler lines divergently selected for abdominal fat content(NEAUHLF)and the Northeast Agricultural University F2 Resource Population(NEAURP)were used,polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)method was developed to detect polymorphisms of ACOX3 gene 5’ Flanking-Region,and associations of polymorphisms with growth and body composition traits were investigated.One mutation named c.-442A>C was found.In the combined populations consisting of 8th to 10th generation of NEAUHLF,c.-442A>C polymorphism was significantly correlated with body weight at 5 and 7 weeks(BW5,BW7),carcass weight(CW),pectoralis minor weight (PMiW),pectoralis minor weight percentage(PMiW/BW),spleen weight(SW),spleen weight percentage (SW/BW)(P<0.05),and suggestively significantly correlated to some extents to body weight at 1 and 3 weeks (BW1,BW3),pectoralis major weight(PMaW),pectoralis major weight percentage(PMaW/BW),leg muscle weight percentage(LMW/BW),heart weight(HW),heart weight percentage(HW)(P<0.2).In the NEAURP, c.-442A>C polymorphism was significantly correlated with birth weight(BWO),body weight at 10-12 weeks (BW10-12),CW,PMiW,PMiW/BW,metatarsus circle of 4 weeks(MeC4),metatarsus length of 6,10 and 12 weeks(MeL6,10,12),femur length(FeL),shank length(ShL),Shank weight(ShW)(P<0.05),and suggestively significantly correlated to some extents to body weight at 8 and 9 weeks(BW8,9),metatarsus length of 8 weeks(MeL8),metatarsus circle of 8 weeks(MeC8),metatarsus claw weight(MeCW),metatarsus claw weight percentage(MeCW/BW).These results indicate that ACOX3 gene extensively affects the growth and body composition traits in chickens.The c.-442A>C polymorphism in ACOX3 gene significantly affects the body weight and muscles weight traits in chickens,and can be applied to molecular MAS programs as a potential marker.
