黄瓜绿斑驳花叶病毒外壳蛋白和运动蛋白基因克隆及序列分析

【摘要】 采用RT-PCR方法和cDNA克隆技术,通过测序获得黄瓜绿斑驳花叶病毒三个株系(CGMMV-GZ,CGMMV-AD,CGMMV-AG)长度为1585bp的序列。序列分析结果表明其包含了完整的外壳蛋白(coat protein,CP)和运动蛋白(movement protein,MP)的核苷酸序列。CP基因序列全长486bp,编码161个氨基酸;MP基因全长795bp,编码264个氨基酸。CGMMV-GZ与GenBank中的两个辽宁株系(CGMMV-LN、CGMMV-Liaoning)外壳蛋白核苷酸组序列一致,运动蛋白核苷酸同源性分别为99.7%和99.5%;CGMMV-AD和CGMMV-AG的外壳蛋白和运动蛋白的同源性均为100%,为同一株系。结合GeneBank中已登录的CGMMV不同序列进行同源性分析,结果显示CGMMV各株系之间外壳蛋白核苷酸同源性分别在90.8%~100%,运动蛋白核苷酸同源性分别在92.7~100%;各分离物和侵染葫芦科植物的小西葫芦花叶病毒(ZGMMV-ZT-1)、kyuri绿斑驳花叶病度(KGMMV-yodo)相比,核苷酸平均同源率54.2%,氨基酸平均同源率为52.2%,差异显著。构建系统发育树显示试验测序的3个分离物与韩国、日本的分离物亲缘关系极近,可能具有共同的侵染源。
【Abstract】 Three RNA nucleotide sequences of 1585bp respectively,of CGMMV-LN,CGMMV-AD, CGMMV-AG isolates were sequenced by cDNA clone generated by RT-PCR.The results revealed that every sequence contains complete coat protein gene and movement protein gene,which are 486 and 795 nucleotides respectively,and encodes the coat protein of 161 amino acids and the movement protein of 264 amino acids.Multiple sequences aligment comparing to other CGMMV isolates which downloaded from GenBank showed that there is no different in coat protein between CGMMV-LiaoN and CGMMV-Liaoning and CGMMV-LN,both of which were designed by other researchers,but a little difference presented in movement protein genome.And the genome of isolates CGMMV-AD and CGMMV-AG is completely identical,it might be the same one.The nucleotide and amino acids identities of coat protein are 90.9%~100%and 94.4%~100%,while movement protein are 93.1~100% and 97.0%~100%among all the tested isolates,respectively.About average 54.2%in nucleotide and 52.2%in amino acid homology were displayed when compared with other cucurbit-infecting tobamoviruses.The results concluded above suggests that there was no marked diversity among the CGMMV isolates,they present very close relatives among each other that from different countries,and might so far as to share the same infected source.
