影响金龟子绿僵菌附着胞形成的理化因子研究

【摘要】 在三种营养液、四种昆虫表皮浸提物、两种虫翅浸取液、三种化学诱导物以及微生态条件下研究了金龟子绿僵菌分生孢子萌发和附着胞形成。绿僵菌分生孢子在0.01%PM、0.012 5%YEM、1%1/4 SDY中附着胞形成率最高。在2.3×10~6~5.3×10~6个/ml的孢子浓度下,接种4 ml 0.012 5%YEM,并在25~26℃、pH 7.0±1.0、相对湿度99%~100%、光照培养条件下最适于附着胞形成。与对照相比,蝉蜕粉与蝇蛆粉浸出液、蝗翅和蝉翅经营养液浸泡灭菌或直接放入营养液均显著促进附着胞形成。不饱和脂肪酸对附着胞的形成有明显促进作用。

【Abstract】 The rate of conidium germination and appressorium formation of Metarhizium anisopliae influenced with three nutrition solutions,extracts of powder of four insect cuticles,wings of two insect species,three chemical inducers and microeconomic environmental factors were determinated. Conidia were inoculated in 0.01%peptone,0.0125%YEM(yeast extract media) and 1%1/4 SDY resulted in the highest appressorium formation rate.With density of conidia(106/ml) and 4 ml 0.01 25%YEM,the optimal culture condition for appressoria formation is 25~26°C,pH 7.0+1.0,RH 99%~100%and slightly lighting.Compared with blank,extract of cicada and powder of fly larva, wings of cicada and locust dipped into 0.01 25%YEM then sterilized or added directly into 0.01 25% YEM facilitated significantly appressorium formation.Unsaturated fatty acid also promoted appressorium development in varied degree.
