生物杀虫剂——昆虫病原异小杆线虫的生物反应器培养

【摘要】 <正>利用计算机控制的15升生物反应器液体单菌培养昆虫病原异小杆线虫Heterorhabditis bacteriophora。培养基经121℃原位高压消毒30分钟。23℃下,接入初生型Photorhabdus luminescens共生细菌,培养48小时后,每ml培养基再接入3000~5000条感染期线虫。生物反应器的装样量10L,温度控制23℃,空气流量为每分钟10L,搅拌速率250~500r/min,自然pH环境。培养12天
【Abstract】 Monoxenic liquid culture of Heterorhabditis bacteriophora H06 was established in 10-liter air lift computer-controlled fermenter.Medium was steam-sterilized in situ for 30 min at 121℃,and inoculated with phase one of Photorhabdus luminescens at 23℃for 2 d.Then dauer juveniles from washed monoxenie cultures were introduced with an inoculum size of about 3000-5000 per ml medium.Culture conditions were.volume in fermenter 10 liters;temperature control 23℃;air flow 10 liters per min;stirring rate 250- 500 r/min;non-controlled pH.Yields of more than 100,000 dauer juveniles per ml were achieved after 12 d,and reached(165±14)×1000 per ml at 18th day,with dauer juvenile proportion of more than 99%.No decrease of the control potential of dauer juveniles produced in fermenter was observed by the standard bioassay method.
