木香中一个新的倍半萜类成分及其生物合成途径分析(英文)

【摘要】 从菊科风毛菊属植物木香(Saussurea lappa)的根中分离得到9个化合物,根据波谱方法鉴定它们的结构分别为:5个倍半萜类成分脱氢木香内酯(dehydrocostus lactone,1),Santamarine(5),β-cyclocostunolide(6),4α-hydroxy-4β-methyldihydrocostol(7)和10α-hydroxyl-artemisinic acid(9);4个其它类成分β-谷甾醇(β-sitosterol,2),胡萝卜苷(daucosterol,3),5-羟甲基糠醛(5-hydroxy-methyl-furaldehyde,4)和trans-syingin(8)。倍半萜及其酯类化合物是风毛菊属植物的特征性成分,在所分离得到的5个倍半萜类化合物中,化合物9是未报道过的新化合物,它是青蒿酸衍生物,其骨架类型在风毛菊属植物中未曾发现,而在菊科蒿属(Artemisia genus)植物中常见。根据分析,推测木香中量最大的成分脱氢木香内酯(1)可能是10α-hydroxyl-artemisinic acid(9)的生物合成前体物质。它在偏弱酸性或相关酶的催化下可转化为10α-hydroxyl-artemisinic acid。
【Abstract】 Five sesquiterpenes,dehydrocostus lactone(1),Santamarine(5),β-cyclocostunolide(6),4α-hydroxy-4β- methyldihydrocostol(7)and 10α-hydroxyl-artemisinic acid(9),along with four other compounds,β-sitosterol(2), daucosterol(3),5-hydroxy-methyl-furaldehyde(4),and trans-syingin(8),were isolated and identified from the radix of Saussurea lappa.Based on the previous reports and our study,sesquiterpene derivatives are the common and characteristic constituents for the genera Saussurea.Among nine compounds obtained,compound 9 is a new sesquiterpene.It is artemisinic acid derivative,whose structure skeleton was not reported from the genera Saussurea(Compositae)before,but artemisinic acid was common compound in another plant of Compositae,Artemisia annua.By the analysis of the compounds in S.lappa,dehydrocostus lactone(1)is a high-content constituent in the plant.Which is a possible bioprecursor of lOa-hydroxyl-artemisinic acid(9).The possible biosynthetic pathway was also speculated.
