中药复方对小鼠骨髓有核细胞及血液生理指标的影响
Effect of Chinese herbal compound on myelosuppressive mice hematopoiesis and immune function

【摘要】 目的:为研制出一种高效低成本的中草药免疫增强剂,并探讨其组方的合理有效性,从血液学角度,研究其对小鼠的造血功能和免疫功能的影响,为免疫增强剂的开发和应用提供参考。方法:(1)取130只6-8周龄、18-22g的清洁级昆明小鼠,按照上述方法复制骨髓抑制小鼠模型,并从中选取100只精神、体况接近的小鼠,并按性别、体重平均分为5组(Ⅰ、Ⅱ、Ⅲ、Ⅳ、Ⅴ组),每组20只(♀♂各半);另取20只健康小鼠(♀♂各半)为Ⅵ组。Ⅰ、Ⅱ、Ⅲ组用中药复方灌胃,剂量(相当于生药含量)分别为:20 g/kg、10 g/kg、5g/kg;Ⅳ组用黄芪多糖,按66 mg/kg灌胃;Ⅴ组为模型对照组、Ⅵ组为健康对照组,用蒸馏水。于每日上午9:00,将制备的中药煎剂按相应剂量用蒸馏水稀释成一定浓度,各组均按0.2 mL/10g灌胃,1次/d,连用3或7次。于用药3d和7d后24h分别采样,每组每次按性别、体重各选取10只小鼠取样。(2)采用血细胞自动分析议测定试验小鼠的骨髓有核细胞(BMC)和血液生理指标,用1%煌焦油蓝盐水溶液染色、显微镜计数网织红细胞(reticulocyte count,Ret)。结果:(1)试验期间,所有小鼠的BMC呈先升高后降低的趋势,其中骨髓抑制小鼠的BMC变化极显著(P<0.01)。用药3d,高剂量组极显著高于模型组(P<0.01);用药7d,健康对照组极显著高于模型组(P<0.01),显著高于高、中剂量中药复方组和黄芪多糖组。(2)各剂量中药复方及黄芪多糖均可升高骨髓抑制小鼠各类白细胞如GRA、LYM和MID,提高GRA%和MID%,降低LYM%;升高小鼠的Ret、RBC、PLT,提高HGB含量。以中药复方10g/kg剂量灌胃7d效果最佳。结论:中药复方具有增强骨髓抑制小鼠造血功能的作用,可以提高骨髓抑制小鼠的BMC、Ret、GRA、LYM、MID、RBC、PLT等,以及血清IL-3和EPO和IL-6的含量;效果以10g/kg剂量,连用7d最佳。
【Abstract】 Objective:To develop an efficient and low-cost Chinese herbal medicine immunopotentiator,the model of myelosuppression mice were established to confirm the efficiency of Chinese herbal compound(CHC ) on hematopoietic and immune from hematology.Method;(1) There were 130 mice as described above were used to establish bone marroif suppression mouse model;and 100 of these mice were selected by spirit,body condition and divided into five groups(Ⅰ,Ⅱ,Ⅲ,Ⅳ,Ⅴ) by gender and quality,20 mice(an equal split of♀and♂) in every group.Another 20 healthy mice(an equal split of♀and♂) were selected for the control group(VI).Mice in groupⅠ,Ⅱ,Ⅲwere orally taken Chinese herbal compound respectiyely in doses(equiyalent to crude drug con- tent) of 20 g/kg,10 g/kg,5 g/kg;group TV were orally taken 66 mg/kg APS;Control groupⅤandⅥwere orally taken distilled water.The traditional Chinese medicine decoction was diluted with distilled water as dose of 0. 2ml(10g body quality) according to the corresponding dose of every group,and 10 mice(an equal split of 9.and 5 ) in a group were giver gavage at 9:00 a.m.one time per day for 3 consecutive days and another 10 mice for 7 consecutive days.Samples were taken at 24h post-gavage the last time.(2) The blood physiological parameters and the mice bone marrow nucleated cells(BMC) were detected by Abacus Junior Vet Hematology Analyzer;and the reticulocytes(Ret) were counted with microscope by stained with 1%brilliant cresyl blue salt.Results:(1) BMC in all mice were first increased and then decreased in the trial period,and BMC in the myelosuppressive mice changed significantly(P <0.01).BMC in group I for orally taken CHC 3 consecutive days were increased significantly than that of the model group(P <0.01);and BMC in healthy control group for orally taken medication 7 consecutive days were increased significantly than that of the model group(P <0.01),and increased significantly than those of in groupⅠ、ⅡandⅣ(P <0.05).(3) CHC groups(group I,group II,group IB) and astragalus polysaccharides could improve various types white blood cells,such as GRA,LYM and MID,and improve the GRA%and MID%,decrease LYM%;and increase Ret,RBC,PLT,increase the content of HGB in the bone marrow suppression mice.The dose of 10g/kg CHC for 7 days had best effectiveness on the bone marrow suppression mice.Conclusion;(1) Chinese herbal compound can improve the hematopoietic function of myelosuppressive mice.The BMC,peripheral blood Ret,GRA,LYM,MID,RBC and PLT of the mice treated with Chinese herbal compound increased.
