清胰化积方下调Ski表达抗胰腺癌生长研究

【摘要】 目的清胰化积方(Qingyihuaji Formula,QYHJ)是我科治疗胰腺癌一个既定、有效方剂,本研究拟探讨QYHJ是否通过下调Ski表达抑制胰腺肿瘤的生长。方法首先通过real-time PCR和western blot对清胰化积方治疗后人胰腺癌裸小鼠皮下移植瘤Ski mRNA和蛋白表达进行检测;然后通过慢病毒介导的RNA干扰技术建立Ski低表达人胰腺癌细胞系,并通过建立裸小鼠皮下移植瘤,评价Ski不同表达肿瘤对QYHJ治疗反应性是否存在差异。结果 QYHJ治疗后人胰腺癌SWl990裸小鼠皮下移植瘤SkimRNA和蛋白表达量明显下降。随后,我们构建了Ski低表达人胰腺癌细胞系SW1 990/Ski RNAi和对照细胞系SWl990/con RNAi,分别建立裸小鼠皮下移植瘤模型,然后给予QYHJ治疗后,发现Ski高表达人胰腺癌细胞(SWl990和SWl 990/con RNAi)应用QYHJ后其抑瘤率为29.6%和32.2%,而Ski低表达人胰腺癌细胞(SWl990/Ski RNAi)应用QYHJ治疗后抑瘤率仅为16.0%一17.8%,Ski下调增加了人胰腺癌细胞对QYHJ治疗的敏感性。结论 Ski为QYHJ治疗胰腺癌一个重要作用靶点,其不同表达介导了胰腺癌细胞对QYHJ治疗的反应性差异。

【Abstract】 Objective To study whether Ski acts as therapeutic target of Qingyihuaji formula(QYHJ) in the treatment of SW1990 pancreatic cancer cell.Methods The expression changes of Ski mRNA and protein in SW1990 pancreatic cancer subcutaneous tumor treated with QYHJ were detected by real-time PCR and western blot.Lentiviral short-hairpin RNA expression vectors targeting Ski were used for creating SW1990 cell with down-regulated expression of Ski.Semi-quantitative RT-PCR and western blot were used for confirming the interfering efficiency.Next,the response of QYHJ treatment on subcutaneous transplanted pancreatic cancer with different Ski expression status was evaluated.Results Expression of Ski mRNA and protein in SW1990 subcutaneous transplanted tumor after treatment of QYHJ decreased 39.6%and 41.3%of that in the untreated respectively.Then stable transfection cells SW1990/Ski RNAi were created and SW1990/con RNAi was used as control.The expression of Ski mRNA and protein in SW1990/con RNAi and SW1990/Ski RNAi were 105%,123%and 46%,30%respectively of that in parental cells.The tumor weight inhibitory rates of QYHJ on subcutaneous transplanted tumors formed by SW1990 or SW1990/con RNAi were 29.6%and 32.2%respectively,while it was 16.0%to 17.8%when the tumors were formed by SW1990/Ski RNAi.Conclusion Ski acts as therapeutic target of QYHJ in the treatment of SW1990 pancreatic cancer cell,and its expression status mediates different response of QYHJ treatment on SW1990 pancreatic cancer cells.
