白术高效液相色谱指纹图谱的建立及种源差异分析

【摘要】 目的:采用高效液相色谱法建立白术的指纹图谱,并对不同产地的白术种源进行成分比较。方法:采用岛津高效液相色谱仪,资生堂Capcell C18柱(4.6×250mm);以乙腈(A)-水(B)为流动相,梯度洗脱程序(A):0~15 min,75%;15~20 min,75%~95%;20~35 min,95%;35~40 min,95%~75%;40~50 min,75%;流速:1mL/min;检测波长220nm。结果:对13个不同产区的白术醇提取物进行了指纹图谱研究,建立了白术的高效液相指纹图谱,确定了9个共有峰,并对其中的内酯Ⅲ、Ⅰ和苍术酮进行了归属;进一步对不同产地的种源进行分析比较。结论:方法简便、快捷,能够用于白术的种源优选及质量控制。
【Abstract】 Objective:To establish the fingerprints of Rhizoma Atractylodis macrocephalae from different origins with High Performance Liquid Chromatography(HPLC)technology.Which is furthermore applied in the quality comparison of plant resource.Method:In this method,C18 column(4.6×250 mm) was used with the mobile phase containing acetonitrile-water for gradient elution,acetonitrile(A):0~15 min,75%;15~20 min,75%~95%;20~35 min,95%;35~40 min,95%~75%;40~50 min,75%.Result:The HPLC fingerprints of Atractylodis macrocephalae from different origins were established and the correlated coefficients of each were calculated.9 common peaks were determined in HPLC chromatogram,three of them were identified as atractylenolideⅢ、Ⅰand atractylon respectively.Conclusion:The method is simple,reproducible and can be used as plant resource selection and quality control of Atractylodis macrocephalae.
