益气药对慢性心力衰竭心气虚证模型大鼠总肌酸激酶活性、肌酸激酶同工酶及腺苷酸转位酶mRNA表达的影响

【摘要】 目的:观察益气药对慢性心力衰竭(CHF)心气虚证大鼠模型总肌酸激酶(CK)活性、CK同工酶及腺苷酸转位酶mRNA表达的影响。方法:结扎大鼠左冠状动脉前降支复制心梗后心衰大鼠模型。按照干预手段分为假手术组(J)、模型组(M)、西药组(卡托普利)(X)、党参组(D)、黄芪组(H)、党参+黄芪组(Y)、联合用药组(党参+黄芪+卡托普利)(L)共7组,连续灌胃9周,酶偶联法检测大鼠血清总CK活性的变化,RT-PCR法检测大鼠心肌CK同工酶和ANT异构体mRNA表达的变化。结果:①CHF模型组大鼠血清总CK活性较假手术组明显降低,各用药组总CK活性均较模型组升高。②模型组CK-B亚基mRNA表达较假手术组明显升高,各用药组的表达均较模型组有显著回落;模型组CK-M亚基mRNA表达较假手术组显著降低,各用药组的表达与模型组相比有明显回升;模型组mi-CK的mRNA表达与假手术组相比无明显差异,西药组表达较假手术及模型组均明显降低,党参组及党参+黄芪组较模型组明显升高。③模型组ANT1、ANT2 mRNA表达较假手术组明显降低,各用药组较模型组均有显著回升。结论:慢性心力衰竭模型大鼠总CK活性、CK同工酶和ANT的mRNA表达水平降低,说明能量代谢尤其是能量物质转运出现异常,在益气药的作用下这些变化有所改善;益气药提高心功能的机制,可能是通过稳定能量物质转运酶系,从而改善心肌舒缩功能。
【Abstract】 Objective:To study the effects of herbs of invigorating qi on CK activity the level of CK isoenzyme and ANT mRNA gene expressions of chronic heart failure(CHF) rats.Methods:Rat models of heart failure were made by ligaturing coronary artery.Divide the rats into 7 groups,which were sham-operation group,CHF model group,Captopril group,Codonopsis group,Astragalus group,Codonopsis & Astragalus group,combination group.Taking the drugs by oral gavage for 9 weeks.Study the effects of herbs of invigorating qi on CK activity the level of CK isoenzyme and ANT mRNA gene expressions by the way of Western blots.Results:①On the 9th week,the CK activity of CHF model group was much lower than that of the sham-operation group(P<0.01),the treatment groups could prevent it from being reduced,of which Codonopsis group,Astragalus group,combination group were significant when being compared with the model group(P<0.05,P<0.01).② The level of CK-B mRNA gene expressions of model group were higher than that of the sham-operation group(P<0.01).The Codonopsis group,Astragalus group,Codonopsis & Astragalus group,combination group could reduce it down obviously(P<0.05,P<0.01).The levels of CK-M mRNA gene expressions of CHF model group were much lower than the sham-operation group(P<0.01).The Codonopsis group,Astragalus group,Codonopsis & Astragalus group,combination group could rise it up obviously(P<0.05,P<0.01).The levels of mi-CK mRNA gene expressions of CHF model group were no different from the sham-operation group.Captopril group was lower than the sham-operation group & CHF model group(P<0.05,P<0.01),Codonopsis group & Combination group was lower than the CHF model group(P<0.05).③ANT 1 & 2 mRNA gene expression of CHF model group were lower than the sham-operation group(P<0.05),all the treatment groups could rise it up,Codonopsis group,Astragalus group,Codonopsis & Astragalus group,combination group could raise the level of ANT mRNA gene expression significantly(P<0.01) compare with the model group(P<0.05,P<0.01).Conclusion:The CHF model rats showed the level of ANT mRNA gene expressions and CK activity of failed cardiomyocytes decreased.These changes showed that dysfunction of energy metabolism especially energy transportation emerged.Herbs of invigorating qi could improve this dysfunction.According to research above,we infer that the mechanism of improving heart function with herbs of invigorating qi may be concern with stabilizing the enzymes used for transporting energy substances so as to improving diastolic and systolic function.
