水通道蛋白-4在缺血/缺氧损伤后神经血管单元中的作用

【摘要】 <正>背景:神经血管单元由神经元、提供营养的微血管和支持细胞如小胶质细胞和周细胞等组成,并强调在不同的神经元、神经胶质细胞和血管腔隙之中的细胞间的信号,这是维系正常脑功能稳态的基础。但在缺血性脑水肿时AQP4在神经血管单元各组成成分中的作用仍然不甚清楚。目的:探讨基于神经血管单元的组成中存在不同的细胞成分,水通道蛋白-4可能在缺血性脑水肿中发挥不同的作用。方法:免疫印迹法检测bEnd.3细胞株水通道蛋白-4的表达;免疫印迹法检测缺血/再灌注大鼠6 h,24 h和48 h在体分离的脑微血管水通道蛋白-4的表达;免疫印迹法检测缺氧缺糖损伤后培养的星形胶质细胞水通道蛋白-4表达。结果:在鼠大脑微血管内皮细胞株bEnd.3观察到水通道蛋白-4的表达。与对照组比较,在大鼠缺血/再灌注组6h,24h和48h分离的脑微血管内皮细胞水通道蛋白-4表达水平增加。与对照组比较,缺氧缺糖损伤组培养的大鼠星形胶质细胞水通道蛋白-4表达下降。结论:存在于脑微血管的水通道蛋白-4可能促进缺血性中风早期脑水肿,神经胶质细胞水通道蛋白-4的暂时丧失可能对缺血后脑水肿的形成具有保护作用。
【Abstract】 A historical and chronological research of ancient Chinese medical treatises revealed that the records of post-stroke epilepsy and its therapy could date back to Eastern Han Dynasty.Synopsis of Prescriptions of the Golden Chamber by Zhang Zhongjing (AD 152-219) has recorded the Tanjian(Paralysis and Epilepsy) under the chapter titled "Apoplexy" which was treated by Feng-yin Decoction.The description of post-stroke epilepsy was confirmed in many others published later.Through nearly two thousands years of transmission and development,the prescription Feng-yin Decoction is still being used today.It has been claimed to be effective in clinical practice and showed antiepileptic and anticonvulsive effects in modern pharmacologic researches.These reports lead to the conclusion that post-stroke epilepsy is first described in about 2000 years ago in China.
