前列活血汤对自身免疫性前列腺炎大鼠模型前列腺内炎性细胞因子的影响

【摘要】 【目的】观察前列活血汤对自身免疫性前列腺炎大鼠模型(EAPAM)前列腺组织内炎性细胞因子的影响,从而探讨中药治疗前列腺炎的作用机制和作用靶点。【方法】60只雄性Wistar大鼠随机分成6组:正常对照(G1)组、模型(G2)组、前列泰(G3)组、前列活血汤(G4)组,每组10只;余两组20只制作前列腺组织抗原用。G2、G3、G4组经腹部皮下多点注射大鼠前列腺组织抗原造模EAPAM。免疫造模3天后,各组动物分别予生理盐水、前列泰、前列活血汤灌胃8周。处死后光镜观察各组大鼠前列腺组织的病理改变;酶联免疫吸附法(ELISA)检测大鼠前列腺组织中白介素8(IL-8)、白介素10(IL-10)、肿瘤坏死因子α(TNF-α)的水平;聚合酶链式反应法(PCR)检测检测大鼠前列腺组织中TNF-αmRNA的表达水平。【结果】模型组前列腺组织中IL-8、IL-10、TNF-α、TNF-αmRNA表达水平较正常对照组均明显升高(P<0.05);前列腺活血汤组前列腺组织中IL-8、IL-10、TNF-α、TNF-αmRNA表达水平较模型组均有所降低(P<0.05)。【结论】前列活血汤能通过调节前列腺局部免疫状态从而明显减轻模型大鼠前列腺的炎症反应。
【Abstract】 [Objective]To explore the acting mechanism and acting target of Qianliehuoxue Decoction(QD) in treating prostatitis by observing it’ s effect on inflammatory factors in autoimmune prostatitis rat mode.[Methods]60 rat were randomized into 6 group:the normal group(G1),the model group(G2),the Qianlietai Tablet(QLT) group(G3),the QD group(G4),10 per group.The other two groups(n=20) were used for making prostate tissue antigen.The rat model(G2, G3,G4) was established by abdominal subcutaneous multiple points injection of rats’ prostate tissue antigen.They were treated via gastrogavage respectively with normal saline,QLT and QD for 8 weeks three days after injection.Pathologic change of prostatic tissue was observed by Optical microscope after All rat were sacrificed.And the levels of interleukin 8(IL-8), interleukin 10(IL-10) and tumornecrosis factorα(TNF-α) in prostatic tissue were detected by enzyme linked immunosorbent assay(ELISA).The expression levels of TNF-αmRNA in prostatic tissue were detected by Polymerase Chain Reaction(PCR).[Results]Compared with the normal group, the prostatic expression levels of in the model group were higher(P<0.05).In the QD group, the prostatic expression levels of IL-8,IL—10,TNF-αand TNF-αmRNA were lower than the model group(P<0.05).[Conclusion]QD could reduce the prostatic inflammatory response of model rats significantly by regulating the local immune condition.
