电针对脑缺血再灌注大鼠神经黏蛋白表达和脑含水量的影响

【摘要】 目的观察电针对易卒中型肾血管性高血压大鼠脑(RHRSP)缺血后再灌注不同时间神经黏蛋白表达、脑组织含水量的干预作用。方法将180只SD大鼠先用环形银夹狭窄双侧肾动脉,制成RHRSP,再用线栓法制成一侧大脑中脉闭塞(MCAO)脑缺血再灌注模型。随机分为假手术组、电针组和对照组,分别观察缺血2h后再灌注1d、7d、14d、30d大鼠神经黏蛋白、脑组织含水量的变化。结果脑缺血再灌注1d,对照组脑缺血区周围出现神经黏蛋阳性表达细胞[(21.17±2.44)个/cm~2],7d明显增多[(33.62±4.39)个/cm~2],14d表达到高峰[(55.17±6.84)个/cm~2],30d下降[(17.83±3.42)个/cm~2]。电针组神经黏蛋白阳性表达细胞在7d[(24.19±3.82)个/cm~2]、14d[(39.78±5.26)个/cm~2],30d[(10.96±2.17)个/cm~2],时较对照组明显减少(P<0.05),脑缺血1、7d,电针组大鼠脑组织含水量(80.72±1.37)、(79.43±1.25)明显小于对照组(83.63±1.33,P<0.001)、(81.65±1.65,P<0.05)。结论电针减轻脑缺血再注损伤后脑水肿,促进缺血损伤区神经功能修复,可能与其下调神经抑制因子神经黏蛋白的表达机制密切相关。

【Abstract】 Subject To observe the effect of electric acupuncture on the expression of Neurocan and cerebral water content at the different time of cerebral ischemia and reperfusion in Stroke-prone-Renovascular-Hypertensive-Rats(RHRSP).Methods After the renal arteries were constricted bilaterally with ring-shape silver clips, the stroke-prone renovasular hypertensive rats(RHRSP) were established in all operated 180 SD rats.The models of focal cerebral ischemia and reperfusion(I/R))of MCAO were created by intraluminal middle cerebral artery occlusion in RHRSP,which were categorized randomly into sham-operated group,electric acupuncture group and control group.The changes of Neurocan and cerebral water content in RHRSP were detected after 2-hour ischemia and 1-day,7-day,14-day and 30-day reperfusion. Results The positive cells of Neurocan were found in ischemic area after 1-day ischemia and reperfusion in control group[n=(21.17±2.44 )/cm~2].They were increased significantly after 7-day reperfusion[n=(33.62±4.39 ) /cm~2]and up to the maximum after 14-day reperfusion[(55.17±6.84 ) /cm~2],but decreased after 30-day reperfusion[n=(17.83±3.42 ) /cm~2].Contrast to the control group,the expression of Neurocan in electric acupuncture group were respectively decreased at 7-day[n= (24.19±3.82) /cm~2],14-day[n=(39.78±5.26 ) /cm~2]and 30-day[n=(10.96±2.17 ) /cm~2].The cerebral water content(80.72±1.37 )、(79.43±1.25) in RHRSP were markedly lower than control group(83.63±1.33 )、(81.65±1.65 )after 1-day and 7-day ischemia, being significantly different(P<0.001,P<0.05)Conclusions The therapeutic action of electric acupuncture on dissipating brain edema to promote the neurofunctional recovery in RHRSP after cerebral ischemic reperfusion injury was related with the down regulation to the expression of Neurocan.
