番荔枝种子中内酯类成分的分离及其主成分细胞毒活性研究

【摘要】 目的:研究番荔枝中抗肿瘤活性成分。观察Squamocin对BGC-823和HL-60细胞的影响。方法:对氯仿提取物用正反相柱,HPLC进行系统的成分分离,光谱解析和结构鉴定。用MTT方法检测Squamocin对体外培养的人癌细胞BGC-823和HL-60细胞的细胞毒性作用。结果:从番荔枝Annona squamosa)种子中分离得到9个白色蜡状固体,其结构分别鉴定为Tripoxyrollin(1),Squamocin(2),Squamocin-B(3),Squamocin-G(4),Moltrilin(5),Squamocin-J(6),Squamocin-L(7),Squamostatin-A(8),Squamostatin-C(9),其中Tripoxyrollin是首次从该植物中分离得到的天然产物。在squamocin浓度1x10-6 and 10μg/ml时,在BGC-823和HL-60细胞第1天就能测到细胞存活率明显降低。结论:本实验从细胞水平证实Squamocin对体外培养的两种人体癌细胞株胃癌(BGC-823)和人急性早幼粒白血病细胞(HL-60)具有很强的杀伤作用,且呈现良好的剂量依赖关系。

【Abstract】 Objective:To study the cytotoxic compounds from Annona Squamosa.Methods:Using chloroform extraction and silica gel column chromatography to isolate compounds and using spectral means to determine the structures. The cytotoxic activity against human cancer cell lines BGC-823 and HL-60 was assayed by MTT methods.Results:Nine compounds were identified as tripoxyrollin(1),squamocin(2),squamocin-B(3),squamocin-G(4),moltrilin(5), squamocin-J(6),squamocin-L(7),squamostatin-A(8) and squamostatin-C(9),respectively.Among them,tripoxyrollin(1) was firstly isolated from Annona squamosa.When BGC-823 and HL-60 cells was treated with squamocin 1×10-6 and 10g/ml for 1 day,respectively,cells survival ability was remarkably inhibited.Conclusion:Squamocin exhibited potent cytotoxic activity in vitro against two human tumor cell lines(BGC-823 and HL-60 cells).
