【摘要】 目的:探讨仙茅、淫羊藿含药血清单用或与TAM合用是否有促人乳腺癌MCF-7细胞增殖/凋亡的作用。方法:设立空白、E2及TAM对照组,观察仙茅、淫羊藿含药血清单用及联合TAM在不同剂量和作用时间下对MCF-7细胞的生长的影响。应用MTT法观察细胞增殖情况,应用Annex-V+PI双染法及流式细胞仪观察细胞凋亡情况。结果:仙茅、淫羊藿对乳腺癌MCF-7细胞生长的影响因浓度的不同而不同:不同浓度的仙茅含药血清对乳腺癌细胞MCF-7的促增殖作用均不明显(P>0.05)。仙茅、淫羊藿与TAM联合作用于MCF-7细胞,无明显促细胞增殖的作用(P>0.05),高剂量的淫羊藿甚至有诱导细胞凋亡的可能(P<0.05)。结论:仙茅、淫羊藿临床上使用基本是安全的,尤其对ER(+)乳腺癌患者合并TAM治疗的情况下。从而为临床应用补肾中药治疗乳腺癌提供了实验依据。更多还原

【Abstract】 Objective: The aim of the study was to explore the effects of Urculigo orchioides and Epimedium serum on the prolife ration and apoptosis of breast cancer MCF-7 cells. Materials and methods: The different doses of Urculigo orchioides and Epimedium serum alone and combined with tamoxifen(TAM) were added into the culture medium,and 17β-estradiol(E2),TAM were also added to culture medium as control groups.The MTT assay was used to evaluate the proliferation of MCF-7 under the different conditions of interventions.Cell apoptotic rate were measured with flow-cytometry and Annexin v-PI staining.Cellular morphologic changes were observed using transmission electron microscopy.Data are expressed as mean±standard deviation(SD),and a one-way ANOVA was performed for each group.All statistical tests were two-sided and p<0.05 was considered to indicate statistical significance. Results: The influences of cell growth were different with the different concentrations of Urculigo orchioides and Epimedium.The stimulatory effect of Urculigo orchioides was weak and there is no significant differences between Urculigo orchioides serum with the blank control(p>0.05). Urculigo orchioides and Epimedium of various concentrations combined with TAM show no effect on the proliferation of breast cancer cells.On the other hand,Urculigo orchioides and Epimedium could cause profound inhibition of growth of MCF-7 at higher doses.20%Urculigo orchioides and Epimedium serum combined with TAM or Epimedium serum alone can induce apoptosis and the effects were dose-dependent. Conclusions These data suggest that Urculigo orchioides and Epimedium serum combined with TAM have no significant effect on the proliferation of MCF-7 cells;the higher dose Epimedium even has the ability to induce apoptosis of breast cancer cells.The results of the study can provide experimental evidence for Urculigo orchioides and Epimedium applied in breast cancer survivors and suggest that Urculigo orchioides and Epimedium are safety to be used in clinic,especially in those women with ER(+) and receiving therapy of TAM.更多还原
